Expression analysis in response to drought stress in soybean: shedding light on the regulation of metabolic pathway genes Genet. Mol. Biol.
Guimarães-Dias,Fábia; Neves-Borges,Anna Cristina; Viana,Antonio Americo Barbosa; Mesquita,Rosilene Oliveira; Romano,Eduardo; Grossi-de-Sá,Maria de Fátima; Nepomuceno,Alexandre Lima; Loureiro,Marcelo Ehlers; Alves-Ferreira,Márcio.
Metabolomics analysis of wild type Arabidopsis thaliana plants, under control and drought stress conditions revealed several metabolic pathways that are induced under water deficit. The metabolic response to drought stress is also associated with ABA dependent and independent pathways, allowing a better understanding of the molecular mechanisms in this model plant. Through combining an in silico approach and gene expression analysis by quantitative real-time PCR, the present work aims at identifying genes of soybean metabolic pathways potentially associated with water deficit. Digital expression patterns of Arabidopsis genes, which were selected based on the basis of literature reports, were evaluated under drought stress condition by Genevestigator. Genes...

The study of tolerance mechanisms for drought stress in soybean is fundamental to the understanding and development of tolerant varieties. Using in silico analysis, four marker genes involved in the classical ABA-dependent and ABA-independent pathways of drought response were identified in the Glycine max genome in the present work. The expression profiles of the marker genes ERD1-like, GmaxRD20A-like, GmaxRD22-like and GmaxRD29B-like were investigated by qPCR in root samples of drought sensitive and tolerant soybean cultivars (BR 16 and Embrapa 48, respectively), submitted to water deficit conditions in hydroponic and pot-based systems. Among the four putative soybean homologs to Arabidopsis genes investigated herein, only GmaxRD29B-like was not regulated...

Identification and characterization of homeobox genes in Eucalyptus Genet. Mol. Biol.
Rocha,Graça Celeste Gomes; Corrêa,Régis Lopes; Borges,Anna Cristina Neves; Sá,Claudio Bustamante Pereira de; Alves-Ferreira,Márcio.
Homeobox genes encode transcriptional factors, usually involved in molecular control of plant developmental patterns. They can be divided into several classes according to conserved sequences within the homeobox region and the presence of specific additional sequences. Based on these conserved sequences, we developed a search procedure to identify possible homeobox genes in the Eucalyptus Genome Sequencing Project Consortium (FORESTs) database. We were able to identify 50 Eucalyptus sequences (EST-contigs) containing the homeodomain sequence. Phylogenetic analysis was applied to this ESTs-contigs and 44 of them were found to have similarities with one of three well-known homeobox classes: Bell, Knox and HD-Zip, and their sub-classes. However, no EST-contig...

Identification of novel and conserved microRNAs in Coffea canephora and Coffea arabica Genet. Mol. Biol.
Loss-Morais,Guilherme; Ferreira,Daniela C.R.; Margis,Rogério; Alves-Ferreira,Márcio; Corrêa,Régis L..
As microRNAs (miRNAs) are important regulators of many biological processes, a series of small RNAomes from plants have been produced in the last decade. However, miRNA data from several groups of plants are still lacking, including some economically important crops. Here microRNAs from Coffea canephora leaves were profiled and 58 unique sequences belonging to 33 families were found, including two novel microRNAs that have never been described before in plants. Some of the microRNA sequences were also identified in Coffea arabica that, together with C. canephora, correspond to the two major sources of coffee production in the world. The targets of almost all miRNAs were also predicted on coffee expressed sequences. This is the first report of novel miRNAs...

Isolation and characterization of a promoter responsive to salt, osmotic and dehydration stresses in soybean Genet. Mol. Biol.
Conforte,Alessandra Jordano; Guimarães-Dias,Fábia; Neves-Borges,Anna Cristina; Bencke-Malato,Marta; Felix-Whipps,Durvalina; Alves-Ferreira,Márcio.
Abstract Drought stress is the main limiting factor of soybean yield. Currently, genetic engineering has been one important tool in the development of drought-tolerant cultivars. A widely used strategy is the fusion of genes that confer tolerance under the control of the CaMV35S constitutive promoter; however, stress-responsive promoters would constitute the best alternative to the generation of drought-tolerant crops. We characterized the promoter of α-galactosidase soybean (GlymaGAL) gene that was previously identified as highly up-regulated by drought stress. The β-glucuronidase (GUS) activity of Arabidopsis transgenic plants bearing 1000- and 2000-bp fragments of the GlymaGAL promoter fused to the uidA gene was evaluated under air-dried, polyethylene...

Unravelling MADS-box gene family in Eucalyptus spp.: a starting point to an understanding of their developmental role in trees Genet. Mol. Biol.
Dias,Beatriz Fonseca de Oliveira; Simões-Araújo,Jean Luiz; Russo,Claudia A.M.; Margis,Rogério; Alves-Ferreira,Márcio.
MADS-box genes encode a family of transcription factors which control diverse developmental processes in flowering plants ranging from root to flower and fruit development. Members of the MADS-box gene family share a highly conserved sequence of approximately 180 nucleotides that encodes a DNA-binding domain. We used bioinformatics tools to investigate the information generated by the Eucalyptus Expressed Sequence Tag (FORESTs) genome project in order to identify and annotate MADS-box genes. The comparative phylogenetic analysis of the Eucalyptus MADS-box genes with Arabidopsis homologues allowed us to group them into one of the well-known subfamilies. Trends in gene expression of these putative Eucalyptus MADS-box genes were investigated by hierarchical...
